چکیده

Purpose: To investigate the effect of micro-ribonucleic acid (miR)-208a on heart failure (HF) in rats through β-catenin pathway. Methods: A total 24 specific pathogen-free female Sprague-Dawley were enrolled and randomly divided into 3 equal groups, namely, control (normal group), model, study group (miR-208a), with 8 each. Echocardiography was utilized to evaluate cardiac function, terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) staining applied examine cardiomyocyte apoptosis. Finally, expression levels interleukin (IL)-6 IL-10 determined using enzymelinked immunosorbent assay (ELISA). Expression matrix metalloproteinases (MMPs) via immunohistochemistry assay, while western blotting used measure β-catenin.Results: The mRNA level miR-208a significantly lower model than (p < 0.05). Cardiac function better other groups Cardiomyocyte apoptosis increased Furthermore, MMPs, IL-6 elevated comparison those 0.05).Conclusion: MiR-208a reduces inflammatory response deposition extracellular HF inhibition signaling pathway, thereby restoring function.

miR-320 regulates inflammation in EAE through interference with TGF-β signaling pathway

Background: MicroRNAs are small noncoding RNAs that regulate gene expression and involve in many cellular and physiological mechanisems. Recent studies have revealed that dysregulation of microRNAs might contribute to autoimmune disorders such as multiple sclerosis. Based on these findings, we examined the potential role of miR-320 isoforms, miR-320-3p and miR-320-5p, in the context of autoimmu...

TGF-β1 enhanced myocardial differentiation through inhibition of the Wnt/β-catenin pathway with rat BMSCs

Objective(s): To investigate and test the hypotheses that TGF-β1 enhanced myocardial differentiation through Wnt/β-catenin pathway with rat bone marrow mesenchymal stem cells (BMSCs).Materials and Methods: Lentiviral vectors carrying the TGF-β1 gene were transduced into rat BMSCs firstly. Then several kinds of experimental methods were u...

Swimming training alleviated insulin resistance through Wnt3a/β-catenin signaling in type 2 diabetic rats

Objective(s): Increasing evidence suggests that regular physical exercise improves type 2 diabetes mellitus (T2DM). However, the potential beneficial effects of swimming on insulin resistance and lipid disorder in T2DM, and its underlying mechanisms remain unclear. Materials and Methods: Rats were fed with high fat diet and given a low dosage of Streptozotocin (STZ) to induce T2DM model, and su...

Aflatoxin B1 Negatively Regulates Wnt/β-Catenin Signaling Pathway through Activating miR-33a

MicroRNAs are known to play an important role in modulating gene expression in various diseases including cancers and cardiovascular disorders, but only a few of them are associated with the pathology of aflatoxin B1 (AFB1), a potent mycotoxin. Here, we discovered a novel regulatory network between AFB1, miR-33a and β-catenin in human carcinoma cells. The level of miR-33a was up-regulated in he...

Therapeutic inhibition of miR-208a improves cardiac function and survival during heart failure.

BACKGROUND Diastolic dysfunction in response to hypertrophy is a major clinical syndrome with few therapeutic options. MicroRNAs act as negative regulators of gene expression by inhibiting translation or promoting degradation of target mRNAs. Previously, we reported that genetic deletion of the cardiac-specific miR-208a prevents pathological cardiac remodeling and upregulation of Myh7 in respon...
